克隆號為H09的IDH1（isocitrate dehydrogenase 1, 異檸檬酸鹽脫氫酶1）鼠單克隆抗體為Dianova公司近期推出的產(chǎn)品之一，此抗體可特異性的檢測腦組織腫瘤中IDH1基因的R132H點突變。IDH1基因的第132密碼子的雜合突變是II 和III級神經(jīng)膠質(zhì)瘤（根據(jù)WHO的腫瘤學分類）的頻發(fā)事件。在大約70％的星形膠質(zhì)細胞瘤和少突神經(jīng)膠質(zhì)細胞瘤可發(fā)現(xiàn)IDH1基因的R132H點突變。

IDH1基因R132H突變在部分腦腫瘤中的頻發(fā)現(xiàn)象使克隆號為H09的IDH1抗體可高靈敏性和高特異性的用于此類腫瘤的鑒別診斷中：如從原發(fā)性膠質(zhì)母細胞瘤中區(qū)分出間變性星形細胞瘤、從毛細胞型星形細胞瘤或室管膜（細胞）瘤中區(qū)分出彌漫性星形細胞瘤（WHO分類II級），也可以用于具有IDH1突變的腫瘤細胞浸潤前沿的與反應性神經(jīng)膠質(zhì)增多癥的區(qū)分。因此，由于其高靈敏性和高特異性，此抗體在腦部腫瘤的分類分級中以及在檢測單一或少量浸潤腫瘤細胞時是非常有用的。

Antibody clone GHH4 has been developed and validated for the detection of murine CD4 in formalin-fixed paraffin-embedded tissue sections (mouse FFPE). Clone GHH4 identifies T helper cells in murine FFPE tissue by IHC.
CD4 is a marker for T helper cells. Besides this subpopulation of mature T cells (helper/inducer), CD4 is also expressed on most thymocytes and weakly on macrophages and dendritic cells.
Antigen recognition and subsequent T-cell activation is controlled by the interaction between TCR (T-cell receptor) and pMHC (peptide-major histocompatibility complexe). T-cell activation is often enhanced by a close cooperation of the TCR with two different co-receptors: CD8 and CD4. CD4 is a nonpolymorphous 55 kD glycoprotein belonging to the immunoglobulin superfamily. It acts as a co-receptor with the TCR during antigen-induced T cell activation and thymic differentiation by binding MHC class II and plays an important role both in T cell development and in optimal functioning of mature T cells. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail.
In humans, CD4 is expressed in the majority of T-cell lymphomas. CD4 plays an important role in the classification of lymphocytes in inflammatory lesions and malignant lymphomas. CD4 serves as HIV receptor on T cells, macrophages and brain and is downregulated by HIV proteins during AIDS progression.

IHC for CD4 in mouse FFPE tissue

Mouse spleen: The monoclonal antibody clone GHH4 specifically stains CD4 in standard FFPE mouse spleen. Incubation with antibody clone GHH4 (10μg/ml, corresponding to a 1:100 dilution) for 1h at RT. Detection with ImmPRESS HRP anti-Rat from Vector Laboratories (Picture courtesy of Chris Bonnas, Synaptic Systems, Göttingen, Germany).
